The station first signed on the air on September 15, 1953, as WARM-TV. It was the third television station in northeastern Pennsylvania behind WBRE-TV (channel 28) and WDAU-TV (channel 62). The call letters stood for its founder, Walter A. Reuben.

The station originally operated as a CBS affiliate but switched to ABC in 1959. In 1975, Times-Shamrock sold WBRE-TV to Newport Television. Newport sold the station to Nexstar in 2011.

WNEP has been broadcasting in high definition since 2002 and currently broadcasts its news programming in HD. The station also operates a news bureau in Harrisburg, Pennsylvania, and has a fleet of satellite trucks.
